Abstract
Background: Uncaria rhynchophylla is a medicine that has been included in the Vietnamese Pharmacopoeia, with the effect of lowering blood pressure, preventing arrhythmia, sedation, and effects on the smooth muscle system1. In the stem section, the branches with hook-shaped thorns of Uncaria rhynchophylla (Uncaria rhynchophylla (Miq) Jack, Rubiaceae) contain the most saponins compared to other parts. Objective: Developing a procedure to measure total saponins in Uncaria rhynchophylla in Sin Ho district, Lai Chau by photometric methods. Methods: Using the ultraviolet absorbance method after extracting the sample and reacting the Rosenthaler coloration of the saponins with the reagents of perchloric acid and vanillin in glacial acetic acid to give a lilac purple color. Results: The linear range was built with oleanolic acid concentrations in the range of 5 - 17,5 μg/ml with linearity r ≈ 1. The analytical method for accuracy and repeatability gave the results consistent with the analytical requirements. RSD values are 1,9% and 1,5%, respectively. Conclusion: The UV-Vis spectrophotometric method has been successfully developed for the quantification of total saponins in Uncaria rhynchophylla medicinal herbs.
